Is the seminal plasma (SP) and its constituents affect decidualizing capacity and transcriptome of Human main endometrial stromal fibroblasts (eSFs)? SP promoting decidualizing of eSFs of women with and without inflammatory disorders (polycystic ovary syndrome (PCOS), endometriosis) in a way that is not mediated by amyloid semen and those associated with the response transcription powerful, including the induction of interleukin ( IL ) – 11 , cytokines important for the SP-induced decidualizing.
Clinical studies have shown that SP may promote implantation, and in vitro studies have shown that SP can promote decidualizing, steroid hormone driven program differentiation of the ESF which is essential for embryo implantation and impaired in women with disorders of PCOS inflammatory and endometriosis. This is a cross-sectional study in volving samples treated with vehicle alone compared to treatment with SP or SP constituents. SP was tested for the ability to promote decidualizing in vitro in eSFs of women with or without PCOS or endometriosis (n = 9). The role of amyloid semen and SP fractionated in mediating these effects and in eliciting changes in transcription in eSFs were studied.
Finally, the role of IL -. 11 , cytokines with a key role in the implantation and decidualizing, rated as a mediator of decidualizing SP-facilitated eSFs and endometrial epithelial cells (EECS) was isolated from a biopsy of the endometrium of women of reproductive age undergoing benign gynecological procedure and maintained in vitro. Tests were performed to assess whether treatment with SP or SP eSFs constituents affect the rate and extent of decidualizing in women with and without inflammatory disorders.

To characterize the response of the endometrium to SP and SP constituents, RNA was isolated from treated or EECS eSFs and analyzed by RNA sequencing (RNAseq). Factors secreted from cells treated conditioned media were analyzed by Luminex and ELISA. Role IL -. 11 in the SP-induced decidualizing assessed through Clustered interspaced regularly repeated palindromic short (CRISPR) / Cas-9-mediated knockout experiments in eSFs primary SP promoted decidualizing both in the absence and presence steroid hormone (P <0.05 compared to vehicle) with the necessary means seminal proteins.
amyloid semen does not promote decidualizing and induced a weak and transcriptomic responses in eSFs secretomic. Instead, fractionated SP enriched for semen microvesicles (MV) decidualizing promoted. IL – 11 is one of the most potent SP-induced gene in eSFs and important for decidualizing SP-facilitated <.
